Course description
Students will learn basic technique of DNA cloning and concept of that throughout the course, and ultimately master basic genetic engineering technique.
Expected Learning
Students should master basic technique of DNA cloning and learn the concept of gene modification throughout the course, and ultimately master basic genetic engineering technique, think over what’s behind in the experimental results and underlying mechanism.
Course schedule
A class:Students will perform following experiment. Digest λ phage-derived DNA with a restriction enzyme, ligate those fragments into a plasmid vector and transform E. coli bacterial cells. Isolate single colonies by streaking a plate. Purify DNA of interest from E. coli competent cells and confirm that exogenous DNA (DNA of interest) was inserted by (i) digestion with restriction enzyme followed by SDS-PAGE, (ii) determination base sequence.
B class:Purify chromosomal DNA from thale-cress and confirm recombination of genes by using PCR analysis. Students will also purify genomic DNA from Bacillus subtilis and determine the purity of DNA.

Text will be provided in class. Students should also prepare ‘Precaution against accidents and first-aid treatment (Kagaku-Dojin)’ in advance.
