Course description
[Attainment target and theme]
At part of “experiment about superstructure of polymer”, Kazuhiro SHIKINAKA and Kenro TOTANI will teach about basic of measurements of structure formed by polymer. At part of “experiment of organic reaction and synthesis (organic chemistry Ⅱ)”, Akiko OKAMOTO and Michiko MATSUMOTO will teach basic technique such as liquid separation, distillation, and recrystallization. Through this experiments, you will learn reaction behavior of organic molecules and how to handle labile chemical species.
[Experiment about superstructure of polymer]
Knowledge about polymer structure is important for understanding of physical/chemical character of polymer. In these experiments, you will perform experiments about microscopy, X-ray diffraction, and light scattering which are major methods to estimate polymer structure. Before experiments, you will present about homework by oral trial.
[Experiment of organic reaction and synthesis (organic chemistry Ⅱ)]
The bonding formation reaction based on chemical characters of carbonyl group is widely used for synthesis of organic molecules and polymer materials. In these experiments, you will perform synthesis reactions by using high electrophilicity of the carbonyl carbon and high nucleophilicity of enolate.
Expected Learning
To understand the organic reaction of carbonyl compounds by comparing with reactivity, structural features, and stability of the carbonyl compounds.
Course schedule

1. Optical characters of polymer: Optical anisotropy and birefringence analysis
a. basal knowledge of optical anisotropy and birefringence
b. Principle of emergence of interference color (birefringence color)
c. How to use and principle of polarizing optical microscope
d. Determination of principal axis and retardation effect by test plate of unknown anisotropic body
e. Orientation of polymer and its estimation (Birefringence measurement of polymer oriented membrane and fiber)
f. Structural observation and estimation of spherulite and liquid crystal by polarizing optical microscope.
2. Diffraction and Scattering
a. Interference and diffraction of light (diffraction of light by slit and hair)
b. Principle of laser and X-ray
c. Estimation of crystal lattice, degree of crystallinity, and orientation based on X-ray diffraction images
d. Static light scattering

[Experiment of organic chemistry Ⅱ]
1. Grignard reaction: Treating of water-sensitive agent, and temperature control on reaction
2. Wittig reaction: SN2 reaction, liquid separation, and column chromatography
3. Reduction of carbonyl compound: Extraction and reaction tracing by TLC
4. Friedel-Crafts reaction: Reaction control, separation and purification of analogues, and analytical methods
5. Synthesis of ε-caprolactam: Beckmann rearrangement, check of purity by melting point measurement, IR
