Course description
Students will learn basic technique of DNA cloning and concept of that throughout the course, and ultimately master basic genetic engineering technique.

Course schedule
A class:Students will perform following experiment. Digest λ phage-derived DNA with a restriction enzyme, ligate those fragments into a plasmid vector and transform E. coli bacterial cells. Isolate single colonies by streaking a plate. Purify DNA of interest from E. coli competent cells and confirm that exogenous DNA (DNA of interest) was inserted by (i) digestion with restriction enzyme followed by SDS-PAGE, (ii) determination base sequence.
B class:Purify chromosomal DNA from thale-cress and confirm recombination of genes by using PCR analysis. Students will also purify genomic DNA from Bacillus subtilis and determine the purity of DNA.
Prerequisites
In addition to the lectures in the class, you are recommended to prepare and review the class using the standard amount of time as specified by the university for each class.

Text will be provided in class. Students should also prepare ‘Precaution against accidents and first-aid treatment (Kagaku-Dojin)’ in advance.
