Course description
Study about the basic technique for handling of microbes, sterile technique, handling technique of experimental animals, methods for culture media preparation, procedure of cell culture, handling techniques of bacteria and virus, methods for bacteria culture, staining and identification and methods of major serological tests for diagnosis of animal infectious diseases. And understand the principal of these techniques include biochemical nature inspection, serological tests (immune response and basic antigen-antibody reaction).
Expected Learning
Acquire the sterile technique, handling technique of experimental animals, methods for culture media preparation, procedure of cell culture, handling techniques of bacteria and virus, methods for bacteria culture, staining and identification and methods of serological tests. Understand the principal of immune response and basic antigen-antibody reaction.

Course schedule
Practice contents:
1) Understanding of aseptic operation and learning of techniques (Taniguchi).
2) Cell culture, passage, virus inoculation, cytopathic effect (CPE) observation (Taniguchi).
3) Detection of anti-bovine herpesvirus type 1 (BHV-1) antibody in blood by enzyme-linked immunosorbent assay (ELISA) (Taniguchi).
4) Immune reaction (slide agglutination, gel sedimentation, test tube agglutination using Salmonella immunized rabbit serum) (Taniguchi).
5) Preparation of normal agar medium, culture of bacteria, observation of colonies, Gram staining (Furuya).
6) Antibiotic susceptibility test (Furuya).
7) Biochemical characterization of intestinal bacteria (Furuya).

Have to be studying or already finish the course of “General Veterinary Microbiology”

Assessment/Grading
Students who take this practice have to turn in the reports every practice topics. Class participation (60%), report (30%) and attitude of tackling to practice (10%).
Message from instructor(s)
Never forget this practice is the training for handling of pathogenic microbes that cause infectious diseases in some cases. Eating and drinking at the practical room during the practice is strictly forbidden. Anyone who left a white robe and scuffs cannot receive the practice. When there are students who misbehave oneself and take a dangerous behavior during this practice, these students have to get out from this practice room.
