Course description
Students practically learn aseptic handling, characteristics and handling of pathogenic agents that is fundamental technique in veterinary medicine. And then, practically learn the basics of diagnoses in microbiology. Learning the characteristics of bacteria and virus might leads students to handle bacteria and virus appropriately, and to perform diagnoses using fundamental immunological methods. The contents of this laboratory class is based on the technique learned in Practice in Veterinary Microbiology I laboratory class.

Practically learn the contents described below.
Course schedule
Contents are performed over several weeks, and several contents are done simultaneously in one day, depending on the duration of growth or proliferation of cells and pathogens. The schedule is changed every year because of the closing of University by University policy for students committee.
Contents are shown below.

1. Significance and technique of diagnoses of infectious diseases.
2. Bacterial isolation from animals.
  ・Staining.
  ・Inoculation of bacteria into animals.
  ・Counting live bacteria(spreading)
  ・Counting live bacteria(evaluation)
  ・Aseptic sampling
  ・Staining of bacteria, and evaluation
3. Complement fixation (CF)
  ・CF(reaction)
  ・CF(evaluation)
4. HA/HI test
  ・Observation of embryonate chicken eggs.
  ・Preparation of virus isolation from specimen.
  ・Inoculation of virus into embryonate chicken eggs.
  ・Daily observation of the injected eggs.
  ・Collection of virus.
  ・HA/HI test
5. Primary culture
  ・Observation of embryonate chicken eggs.
  ・Preparation of chicken fibroblast from embryonate chicken eggs.
  ・Observation of the cells.
6. Neutralize tests
  ・Thawing of frozen culture cells
  ・Passage of culture cells.
  ・Virus inoculation into cells.
  ・Collection of virus.
  ・Storage of virus.
  ・Preparation of cells for assay, and passage of cells.
  ・Measurement of virus titre(reaction).
  ・Measurement of virus titre(evaluation).
  ・Neutralize test(reaction).
  ・Neutralize test(evaluation).
  ・Storage of cells.
7. Observation of inclusion body
  ・Observation of inclusion body by Giemsa staining.
8. Indirect fluorescence microscopy
  ・Mono-color Indirect fluorescence microscopy
  ・Double staining of Indirect fluorescence microscopy

Prerequisites
Students must have completed the lecture of Veterinary Microbiology and Laboratory class of Practice in Veterinary Microbiology I.

Assessment/Grading
Students are graded by attitude of attendance (60%), report (30%), and examination(10%).
Student will not be able to take the examination nor to get evaluation when the student exceed the number of absences specified in the University policy. Late arrival is treated as same as absence.
Message from instructor(s)
Students handle pathogenic microorganisms in biosafety level 2 facility (BSL2). It's strictly prohibited to eating, smoking nor making up in BSL2 facilities. Students without white coat and slippers for BSL2 facility could not attend laboratory class. Students who is inappropriate attitude, especially making other students involved in danger, must go out of BSL2 facilities.
