Course description |
The purpose of this lecture is to teach students 1) how to purify organic compounds and 2) how to identify organic compounds from spectral data. |
Expected Learning |
To understand how to purify organic compounds. To acquire the ability to deduce the structure of organic compounds from various \spectral data. Corresponding criteria in the Diploma policy: See the curriculum maps. https://www.tuat.ac.jp/campuslife_career/campuslife/policy/ |
Course schedule |
The instrumental analysis methods discussed in this lecture are for purification and separation (GC and HPLC) and for structural analysis (UV, IR, NMR, and MS). 1. introduction 2. Chromatography: (1) The principle and characteristics of separation of GC, HPLC, etc. for purifying and isolating organic compounds will be described. 3. Ultraviolet (UV) absorption spectrum: The theory and characteristic absorption of this method that can obtain information on a conjugated system will be described. 4. Infrared (RI) absorption spectrum: The measurement method of the present method for examining the functional group of a compound, and the features of its structure and spectrum will be described. 5. Mass (MS) spectrum: The principle of measurement and the apparatus, the method of analyzing spectra, various measuring methods, and the like of the present method for providing information on the molecular weight and partial structure of a compound will be described. 6. Nuclear Magnetic Resonance (NMR) Spectrum-1 (Overview, 1H NMR): Measurement of 1H (proton) and 13C nuclear magnetic resonance methods that give information on the bonding state between hydrogen atoms and carbon atoms that constitute the skeleton of an organic compound. The principle, spectrum analysis method, various measurement methods, and information obtained thereby will be described. Since a large amount of information can be obtained in instrumental analysis and is the main method for determining the structure by instrumental analysis, a sufficient time will be described. 7. Nuclear magnetic resonance (NMR) spectrum-2 (13C NMR) 8. Nuclear magnetic resonance (NMR) spectrum-3 (2D NMR) 9. Nuclear magnetic resonance (NMR) spectrum-4 (exercise) Ten. Nuclear magnetic resonance (NMR) spectrum-4 (Explanation and review of exercises) 11. Absolute configuration (optical rotation, ORD, CD, method of determining absolute configuration, stereochemistry and biological activity) 12. Structural analysis using various spectra 13. Structural analysis using various spectra (practice) 14. Structural analysis using various spectra (analysis and review of exercises) 15. Summary, final exam |
